If and when you decide to tell them (try not to pick one of their birthdays, a major holiday, or a particularly stressful workday... the heightened euphoria from the day's events might cause them to react in ways they otherwise wouldn't), sit them down and tell them that there's something very important you need to tell them. When the worried looks on their faces surface, assure them that you are not pregnant, that you are doing fine in school, and that they haven't done anything to make you upset. Then pour your heart out. While you need to be prepared for the possibility of an emotional outburst from one or both parents, remember that they love you, and that being honest with them about yourself is far better than hiding from them forever.
Is your best friend also a lesbian? I'm going to assume that she already knows that you are. I personally don't think it's wise for you to sleep with your best friend, but if you'd like to breach the subject, perhaps the most effective way to do it would be to compliment her on her looks one day, and then half-jokingly say something like, "Man, I would so hook up with you if I could."
